The UVa Transgenic Methods and Applications Workshop
The UVa Gene Targeting and Transgenic Facility conducts a hands-on five day internsive workshop. This workshop includes lectures and hands-on laboratory exercises which is intended for those who wish to develop a strong background in transgenic technologies and applications. On-line registration is available and the cost is $480. Each participant receives a binder containing all materials presented in the workshop along with laboratory protocols and references. The course covers the following areas.
Transgenic technology overview
Early mouse embryo development
Optimizing constructs for in vivo expression
BAC and targeted transgenesis (Rosa26 knock-in)
Making transgenic mice by zygote injection
Preparation of females for embryo collection
Pronuclear microinjection
Generation of pseudopregnant females
Implantation in foster mothers
Identification of transgenic progeny
Homologous recombination in ES cells
Knockout and knock-in construct design
Culture and transfection of ES cells
Generation of gene targeted ES cell clones
Blastocyst microinjection
Optimization of chimeric mouse breeding and germline transmission
Genetic background and speed congenics
