After you are notified by GTTF of the cryo start date, you should initiate the animal transfer by filling out a protocol transfer form(http://www.healthsystem.virginia.edu/internet/ccm/docs/P2.pdf) and submit it to CCM. The male mice should be transferred to GTTF's mouse room MR5 G337. They are used as studs for breeding to donor females to generate embryos for cryopreservation. Please transfer the males only to our protocol # 3520 but keep them on your per diem fund code (CCM #). Please also indicate below whether the males should be euthanized or returned to you after we complete your project.
The donor females are usually wildtype for the allele of interest and ordered by GTTF. In case the females are provided by labs, the required number of females would depend on not only the number of frozen embryos desired but also how well they respond to superovulation. The rule of thumb is that one female produces 5-10 freezable embryos. If requesting to freeze 300 embyros, at least 30 females need to be submitted in several groups. Quality control is assured by thawing and culturing a representative sample of the frozen embryos to assess their ability to develop in vitro.
