Research Description My laboratory conducts basic science research on the involvement of growth factors in cancer. Although dysregulation of growth factor systems has been implicated in a variety of cancers, our primary focus is squamous cell carcinoma of the head and neck, which affects the mucosal surfaces of the upper aerodigestive tract. Our main goals involve: (1) identifying the mechanism by which the insulin-like growth factor-1 receptor (IGF-1R) and other growth factor receptors can confer resistance to targeted therapy with anti-epidermal growth factor receptor (EGFR) agents; (2) determining the benefit of inhibiting additional growth factor receptors and/or downstream signaling molecules to increase the therapeutic efficacy of targeted anti-EGFR agents; and (3) discovering methods to predict the susceptibility of a given tumor to targeted therapy with anti-EGFR agents or with combinations of multiple targeted agents. To address these issues, we study cell lines, xenograft models, and human tumor samples using a variety of current and emerging methodologies.
