Printed Microarrays

The Biomolecular Research Facility offers low-density DNA microarrays printed with gene specific oligonucleotide probes typically ranging from 50-60 bases.   DNA sequences optimized for standard hybridization conditions are selected by using Array Designer 2.0 software (Premier Biosoft International, Palo Alto , CA ).  The oligonucleotide probes are then synthesized on an ABI 3900 DNA Synthesizer (Applied Biosystems, Foster City , CA ).  Once the panel of probes, has been assembled, the probes are printed on Corning UltraGAPS coated slides using an Affymetrix 417 Arrayer (Affimetrix, Santa Clara , CA ).  The printed slides are quality controlled by hybridization with a Cy3 labeled universal oligonucleotide probe. 

 

 

DNA microarray assays gene expression by co-hybridization of two fluorescently labeled cDNA targets, prepared from different samples.   We require approximately 15 µg of total RNA per sample for first strand synthesis of cDNA containing amino-allyl-labeled nucleotides.  The newly synthesized cDNA is then labeled by a covalent coupling of appropriate Cyanine fluor.  RNeasy Kit (QIAGEN, Valencia , CA ) is recommended for standard high quality total RNA purification.

 Mouse cytokines array
This chip with 128 oligo probes in two replicates covers all published mouse cytokine/chemokine ligands and receptors, tumor necrosis factors, and interleukins.
The cost of one slide is $81.00

Human cytokines array

The human cytokine slide represents 164 cytokine/chemokine ligands and receptors, tumor necrosis factors, and interleukins.  Specific probes for 32 G-proteins are also included in the chip.  The probes are arranged in four identical 6x28 spots subarrays, each printed with different pin.  The gene expression level is calculated as an average fluorescence from the four spot replicates.
