Persistent oxidative stress to the lining of small resistance vessels and capillaries stimulates the proliferation of endothelial cells that contribute to occlusive vascular disease. The focus of the work that will be presented links oxidant-induced PTEN and caveolin-1 degradation, eNOS hyperactivation, and defective Notch signaling to endothelial cell dedifferentiation, unproductive angiogenesis, and vascular perfusion defects. Patient-derived tissue sections and cultured endothelial cells together with knockout mouse studies suggest caveolin-1 is an important regulator of endothelial cell-fate determination. - URL:
